Nextera

Illumina's DesignStudio (DesignStudio.illumina.com) supports Nextera Enrichment, a method suitable for larger targeted regions from 500 Kb up to 25 Mb. This targeting method relies upon designing probes that will hybridize to targeted regions allowing selective capture and subsequent sequencing of the regions of interest. In addition to the available Nextera exome enrichment options, DesignStudio allows for custom enrichment.

Nextera Rapid Capture Exome Enrichment

Nextera offers a quick and effective method for targeted enrichment of genomic DNA samples. Using a carefully designed set of probes, Nextera exome enrichment provides high enrichment rates, low duplicates, and exceptional coverage uniformity. With as little as 4 GB sequencing output, over 214K coding exons (37 Mb targeted) are enriched and sequenced, covering up to 98% of the exome (refseq database). Over 80% of the targeted bases are covered at > 10x assuming 4 GB sequencing output.

Nextera Custom Enrichment

Using DesignStudio, we can create custom panels targeting 500 Kb up to 25Mb to your specifications. This probe-based enrichment, requiring only 50 ng of starting material, provides high quality data with > 80% of bases covered at 0.2x the mean read depth. Design your panels yourself or contact your Retrogen Project Manager for assistance in optimizing your designs.

Multiplexing will lower the number of reads per sample and is not recommended for exome sequencing on the MiSeq.
For Exome enrichment, turnaround time is 2 weeks. Custom panels will take longer for panel design, ordering, and shipping.
